Effects of the physico-chemical factors on the lipoxygenase oxidation of linoleic acid

Abstract: Oxidation of linoleic acid by 5-lipoxygenase from potato tubers was studied at different pH-values in the presence of the non-ionic surfactant Lubrol PX, which facilitates UV-monitoring of the process. It is shown that linoleic acid and Lubrol PX form mixed micelles, containing linoleic acid in two major forms with pKal=6.5 (monomeric form localized in the surface layer) and pKa2=8.5 (dimers localized inside hydrophobic core of the micelle). It explains the rise of pH-optimum of the linoleic acid lipoxygenation upon increase of the initial substrate concentration.
