3-CYANO-4-METHYLUMBELLIFERYL-β- D -GALACTOPYRANOSIDE, A NEW CHROMOGENIC AND FLUORIGENIC SUBSTRATE FOR THE β-GALACTOSIDASE FROM E.COLI

Abstract: A novel substrate, 3-cyano-4-trifluoromethylumbelliferryl-β-D-galactopyranoside, for the continuous (pK a = 6,0 for the measured product), photometric (e= 42000 M-1 cm-3 at 440 nm, pH 7,5) and fluorimetric (λex= 440 nm, λem=500 nm) assay of β-galactosidase was synthesized. The substrates affinity to nitrocellulose membranes made possible its use for sensitive (up to 0,1 mole of the enzyme in 510 min) visual identificatioa of the β-galactosidase activity on nitrocellulose sheets.
