DIOLOPHOSPHOLIPIDS. I. SYNTHESIS OF PHOSPHATIDYLHOMOCHOLINES AND BISPHOSPHATIDIC ACIDS ANALOGUES

Abstract: A new method of obtaining diol analogues of phosphatidylhomocholines and bis-phosphatidil acids from easily available 2-oxo-2-(3-hydroxypropyloxy)-1,3,2-dioxaphos-phorinane (the latter is synthesised from 1,3-propandiol by monophoshorylation with cyclic phosphites) has beon developed. Fatty carboxylic acid chlorides easily acylate the free hydroxyl group of oxyalkylcyclophosphate in the presence of Lewis salts. The obtained acyloxyalkyldioxaphosphorinanes alkylates trimethylamine to form diolopho-sphohomocholine and potassium stearate to form bis(propyleneglycol)phosphatidic acid.
