An efficient variant of the solid phase H-phosphonate synthesis of oligodeoxyribonucleotides

Abstract: An efficient variant of the H-phosphonate method of the oligonucleotido synthesis is described. It is based on the use of acetonitrile – quinoline (4:1) mixture as the condensation reaction medium, where the rate of the bis-acylphosphite formation is significantly lower than in pyridine mixtures, thus eliminating the negative effect of preacti-vation of the P-component on the synthesis efficiency. Pentadecamer (Tp)14T was obtained in a high yield at the Victoria-4M automatic synthesizer using a Silochrom S-80 based polymer support.
